CTCF contributes in a critical way to spermatogenesis and male fertility

چکیده
The CCCTC-binding factor (CTCF) is an architectural protein that governs chromatin organization and gene expression in somatic cells. Here, we show that CTCF regulates chromatin compaction necessary for packaging of the paternal genome into mature sperm. Inactivation of Ctcf in male germ cells in mice (Ctcf-cKO mice) resulted in impaired spermiogenesis and infertility. Residual spermatozoa in Ctcf-cKO mice displayed abnormal head morphology, aberrant chromatin compaction, impaired protamine 1 incorporation into chromatin and accelerated histone depletion. Thus, CTCF regulates chromatin organization during spermiogenesis, contributing to the functional organization of mature sperm.

I-4: Male Fertility Preservation Options, Current State-of-The ART and Future Implications
Background Male infertility management has made significant progress during the past three decades, especially after the introduction of intracytoplasmic sperm injection (ICSI) in 1992. However, many boys and men still suffer from primary testicular failure due to acquired or genetic causes. New and novel treatments are needed to address these issues. Spermatogenesis originates from spermatogon...

Background: Cyclophosphamide as an alkalizing and cytotoxic agent contributes in treatment of different kinds of cancer. It can pass the blood-brain barrier. Linking two strains of DNA in cell nucleus and breaking the inner and outer strain linkages and inhibiting RNA synthesis and as a consequence, inhibiting protein synthesis, cyclophosphamide demonstrates severe pharmacological side effects ...
متن کاملThyroid Hormones in Male Reproduction and Fertility
Thyroid hormones, previously thought not to affect spermatogenesis and male fertility, are now being recognized as having important role in spermatogenesis. The identification of thyroid hormone receptors on Sertoli cells, the nurturing cells for sperm in the testis, has embarked further research to investigate the role of thyroid hormones in male reproduction. Since spermatogenesis occurs in c...

Background Imatinib is used in chronic myelogenous leukemia (CML), gastrointestinal stromal tumors (GISTs) and a number of other malignancies. The major aim of this study was to investigate the effects of Imatinib on male fertility in Wistar rats. MaterialsAndMethods Three groups of rats were gavaged with 6, 9, and 12 mg/kg Imatinib dissolved in dH2O for 30 consecutive days. On days 7, 14 and 3...
متن کاملPmn-16: The Effect of Environmental Contaminants on Testicular Function
Male reproductive health has deteriorated considerably in the last few decades. Nutritional, socioeconomic, lifestyle and environmental factors (among others) have been attributed to compromising male reproductive health. In recent years, a large volume of evidence has accumulated that suggests that the trend of decreasing male fertility (in terms of sperm count, quality and other changes in ma...
